KINETIKA PERTUMBUHAN BAKTERI BACILLUS sp. DALAM MEDIUM GLUKOSA SEBAGAI SUMBER KARBON DAN AMONIUM SULFAT SEBAGAI SUMBER NITROGEN

Abstract

Carbon and nitrogen compounds are macro nutrients for bacterial growth. Nevertheless, its high concentration is potential to inhibit bacterial growth. The objective of this research is to study the effects of gucose and ammonium concentrations to the growth of Bacillus sp. producing polyhydroxybutyrate. The experiments was conducted in shaken flask of 250 mL with working volume of 70 mL. Fermentation was incubated in incubator shaker at environmental temperature, initial pH of 7.0, and shaking speed of 130 rpm. The best growth was happened at glucose concentration of 10 g/L and ammonium concentration of 1.4 g/L. The kinetics constants are μm of 0.148 hour-1, Ks (glucose) of 0.413 g/L, and Ks (ammonium) of 0.038 g/L. At high concentration, glucose and ammonium tended to inhibit growth of the bacteria by different pattern. The inhibition constants of glucose and ammonium are 291.022 g/L and 32.913 g/L respectively.
