Frekuensi Alel Minor Gen Transporter Metformin: SLC22A1 rs628031A>G pada Subyek Sehat Indonesia

Abstract


Gen pengkode transporter OCT1, SLC22A1, menunjukkan polimorfisme yang tinggi di populasi Bangsa Asia. Polimorfisme gen tersebut diduga berkontribusi pada variabilitas inter-individual farmakokinetik metformin dengan konsekuensi pada perubahan respon terapi. Di Indonesia, identifikasi polimorfisme gen SLC22A1 sudah pernah dilakukan pada pasien kanker dan diabetes melitus tipe 2, namun pada subyek sehat hingga saat ini belum tersedia data. Penelitian ini bertujuan menentukan distribusi frekuensi alel SLC22A1 rs628031 (A>G) pada subyek sehat Indonesia. Penelitian menggunakan pendekatan deskriptif observasional yang melibatkan subyek sehat Indonesia suku Jawa. Identifikasi SNP menggunakan metode PCR-RFLP. Penelitian menunjukkan bahwa frekuensi alel G gen SLC22A1 rs628031 ditemukan sebesar 62,5%. Kesimpulan, frekuensi alel minor SLC22A1 rs628031 pada subyek sehat Indonesia suku Jawa termasuk tinggi (65,2%), hampir sama dengan frekuensi alel minor pada populasi Bangsa Asia lainnya.
